Research Interest
This research group is responsible for the management of the Gatersleben genebank collection, which entails the long-term storage, multiplication and distribution of the germplasm. Its major research foci concern seed longevity, genetic integrity and genetic diversity, and it also carries out an extensive programme of germplasm evaluation and genetic characterisation. The experimental work is done in close co-operation with other IPK research groups with expertise in molecular and biochemical techniques (Rg Gene und Genome Mapping, Rg Experimental Taxonomy, Rg Applied Biochemistry), as well as with numerous non-IPK institutions (see references below).
 
 
Management
The Gatersleben seed store conserves about 130,000 accessions, held in five cold chambers which are maintained at -15ºC. Every year germination tests are carried out on about 12,000 accessions, and over 20,000 samples are distributed world-wide for research and breeding purposes. Multiplication is required for some 10,000 accessions per year. About 1,500 entries are maintained as living plants in vivo. A quality management system was established (ISO9001:2008 certification), a measure which was taken to improve internal management practices. The full documentation of the management protocols ensures that the long-standing experience and knowledge of current employees is captured for the benefit of their successors.

Seed longevity
Since more than 90% of genebank accessions globally are stored in the form of seed, seed longevity is of particular importance for crop germplasm preservation. At the IPK, research has been initiated for a range of crop species. We have detected variation for this trait both between crop species, and between accessions of the same species. For the genetic analysis of seed longevity, we have applied accelerated ageing tests based on International Seed Testing Association (ISTA) protocols. In addition to exploiting conventional bi-parental mapping populations, we have also been exploring the value of association genetics approaches, which can be used to compare seed lots which have been stored over varying lengths of time (‘natural ageing’ panels). In addition to these genetic studies, seed material is also used for various physiological and biochemical surveys (in co-operation with the Royal Botanic Gardens, Kew, UK and the National Center for Genetic Resources Preservation, Fort Collins, USA).

Genetic integrity
A particular challenge for ex situ genebanks is the maintenance of genetic integrity. Contamination with non-self pollen or inadequate handling during multiplication will inevitably downgrade genetic identity. Molecular markers are used at IPK to control this potential problem by maintaining both the seed harvested from the most recent multiplication and a herbarium voucher specimen collected from the initial multiplication. The cereal herbarium collection includes samples of both the grain and intact spikes of each accession. Pilot DNA fingerprinting studies have revealed that in self-pollinating species such as wheat (Triticum), a high degree of identity is preserved, whereas in open pollinating species such as rye (Secale), allele frequencies can shift over time.

Genetic diversity
Concern has been expressed that urbanization, the replacement of traditional agricultural systems by modern industrial methods and the introduction of modern high-yielding varieties together are putting pressure on genetic diversity. At the IPK genebank materials from several repeat collection expeditions to certain geographical regions are available; these allow for an evaluation of the flux in genetic diversity over a 40 to 50 year period. Whereas quantitative changes are typically small a qualitative allelic flow is generally detectable.

Evaluation and genetic characterisation
In close co-operation with IPK colleagues and external partners, the collection has been widely characterized and evaluated. In the cereals (wheat, barley and rye), a number of bi-parental mapping populations and association mapping panels have been established to allow for the genetic analysis of various traits. The current focus covers tolerance to a number of abiotic stresses, in particular drought, soil salinity and soil soluble aluminium.

Characterization and evaluation activities are also directed at Papaver, Petroselinum, Salvia and Coriandrum, a range of leafy vegetables (Lactuca, Spinacia, Cichorium, Valerianella, Eruca, Diplotaxis) and other genera/species of the collection.
