Mission

Ziel der Ag Angewandte Biochemie ist die Aufklärung molekularer Mechanismen der Regulation des pflanzlichen Sekundärmetabolismus. Von besonderem Interesse ist die Einbettung der Stoffwechselwege in den Gesamtmetabolismus und in die Mechanismen der Stressabwehr; somit tragen die Arbeiten zu den Forschungsschwerpunkten Wachstum und Stoffwechsel (FSP 4) sowie Dynamik der Pflanze-Umwelt-Interaktionen (FSP 5) bei. Neben den pflanzenspezifischen Funktionen bestimmen die Muster an sekundären Inhaltsstoffen auch die Qualität von Futter- und Nahrungsmitteln, da einzelne Komponenten erwünschte Eigenschaften wie anti-oxidative Wirkung, aber auch toxische Effekte zeigen können.

Das Verständnis regulatorischer Prozesse bildet die Grundlage für die gezielte biotechnologische Produktion von wertvollen Inhaltsstoffen und für die Pflanzenzüchtung, um bestimmte Metabolitmuster genetisch festzulegen. Dabei spielt die Verknüpfung mit dem Primärmetabolismus eine wichtige Rolle, insbesondere auch mit dem Nährstoff-Zustand (Kooperation Ag MPE). Weiterhin wird die räumliche und zeitliche Verteilung von Metaboliten mit Hilfe von Massenspektrometrie-basierten und mikroskopisch-zellbiologischen Methoden (Kooperation Ag SZB) untersucht, um zelluläre Netzwerke und Entwicklungsprozesse besser modellieren zu können. Zu diesem Ziel tragen auch gerichtete und ungerichtete Proteomanalysen bei.

Projekte

Ein Schwerpunkt der Arbeiten ist gegenwärtig die Analyse des Phenylpropanstoffwechsels in Gerste. Im Projekt „Barley Biodiversity“ wurden in Zusammenarbeit mit der Universität Halle (Prof. Pillen) eine Vielzahl an Genotypen der Gersten-Populationen HEB-25 und S42 auf ihre Muster an Phenylpropanoiden untersucht. Die phytochemischen Profile wurden für die Kalibrierung von Modellen verwendet, die aus hyperspektralen Daten abgeleitet wurden. Die Modelle sollen verwendet werden, um mit Hilfe der nicht-invasiv erhobenen spektralen Signaturen Voraussagen über Inhaltsstoffe machen zu können. Die phytochemischen Daten wurden von uns auch genutzt, um auf der Grundlage der am IPK vorhandenen genomischer Information neue Enzyme des Phenylpropanstoffwechsels zu identifizieren (Brauch et al., 2018). Eine vertiefte Auswertung der komplexen Datensätze erfolgt in Zusammenarbeit mit der Ag NAM. Weiterführende Experimente sollen die Bedeutung spezifischer phytochemischer Profile für die biotische und abiotische Stressabwehr (Kälte) aufklären.

Die Charakterisierung der Stress-Resistenz (Trockenheit, Stickstoff-Mangel) von kontrastierenden Kartoffel-Genotypen erfolgt mit Hilfe von Metabolit- und Proteomuntersuchungen (BMEL Projekt Valprokar). Im Vorläufer-Projekt wurde eine Reihe von Kandidatenproteinen identifiziert, die nun funktionell charakterisiert werden. Im Vordergrund stehen zwei Aquaporine, die über die Proteomanalyse von Plasmamembran-Fraktionen aus Kartoffelwurzeln identifiziert wurden.  Die funktionelle Charakterisierung wird mit Hilfe von Arabidopsis-Mutanten durchgeführt; die Untersuchung von Transporteigenschaften erfolgt im Hefesystem (Kooperation MPE und Patrick Bienert, München).

Metabolische Modelle werden genutzt, um die Antherenkulturtauglichkeit von Weizen-Genotypen vorherzusagen (INPUT-Projekt mit der Saaten-union Biotec GmbH). Dabei kommen gerichtete Untersuchungen und ungerichtete Analysen durch LC-MS zum Einsatz, um die Daten für die prädiktive Modellierung zu erzeugen.

Metabolische Daten werden ebenfalls genutzt, um die Lagerfähigkeit von Zuckerrüben-Genotypen zu vergleichen (Projekt Beetstore). Die Metabolitdaten werden mit Transkriptdaten in Beziehung gesetzt, um die molekularen Grundlagen für eine gute Lagerfähigkeit des Rübenkörpers aufzuklären (Kooperation mit Dr. Eva Sehr, AIT Tulln). Weitergehende Untersuchen werden durchgeführt, um Kältetoleranz-Mechanismen in frühen Entwicklungsstadien der Zuckerrübe aufzuklären. Zum Einsatz kommen dabei kontrastierende Genotypen aus der Kollektion der IPK Genbank.
