Mission

Mikroskopische Untersuchungen sind ein wesentlicher Bestandteil der modernen Pflanzenforschung. Neben den klassischen morphologischen und strukturellen Aspekten, wurde in den letzten Jahren ein verstärkter Fokus sowohl auf das dynamische Wachstum der Pflanzen als auch auf die Visualisierung zelldynamischer Prozesse gelegt. Vor allem Untersuchungen lebender Pflanzen auf zellulärer und subzellulärer Ebene haben hierbei zunehmend an Bedeutung gewonnen.

Eine der wichtigsten Fragen in der Kulturpflanzenforschung ist derzeit, wie sich die Pflanzen an immer extremere Umweltbedingungen anpassen und welche Strategien hierbei entwickelt werden um mit ihnen umzugehen. Eine wichtige Aufgabe der Arbeitsgruppe Strukturelle Zellbiologie in diesem Bereich, ist die funktionelle Charakterisierung wichtiger Strukturmerkmale mit modernen zellbiologischen und mikroskopischen Methoden. Das auf diese Weise gewonnene Wissen, kann in der klassischen Züchtung und durch die Anwendung neuer Züchtungstechnologien, zu einer gezielten Verbesserung der Pflanzenleistung beitragen.

Im Laufe der Jahre konnte in der Arbeitsgruppe eine moderne zentrale Einrichtung für Licht- und Elektronenmikroskopie am IPK etabliert werden. Zuletzt wurde die umfassende Mikroskopieplattform, zudem durch ein superauflösendes Lichtmikroskop und ein Lichtscheibenmikroskop in der Arbeitsgruppe Chromosomenstruktur und -funktion des IPK erweitert.

Neben der eigenen Forschung sind wir für die Wartung und den geführten Einsatz von Geräten sowie für die Entwicklung neuer Protokolle zur Aufbereitung von Pflanzengewebe für die Bildgebung lebender Zellen, Ultrastrukturanalysen, 3D-Rekonstruktionen und Immunolokalisierungsstudien verantwortlich. Der kombinierte Einsatz der beiden Disziplinen, wird zweifellos die Möglichkeit eines interdisziplinären mikroskopischen Ansatzes zur Beantwortung wissenschaftlicher Fragen erhöhen.

In vielen Fällen sind mikroskopische Untersuchungen Teil eines Projekts, das in Zusammenarbeit mit anderen Forschungsgruppen des IPK oder externen Forschungseinrichtungen durchgeführt wird. Das übergeordnete Forschungsziel eigener Projekte mit den Schwerpunkten Ausfallfestigkeit von Samen, Lagerresistenz und Trockenstress besteht darin, agronomisch relevante Merkmale in Kulturpflanzen zu identifizieren und zu charakterisieren.

Projekte

Das Hauptziel des DFG Gemeinschaftsprojektes mit der AG Pflanzliche Reproduktionsbiologie und der Abteilung Genetik der Universität Silesia, besteht in einer tiefgreifenden Untersuchung zur ABA-SL-Wechselwirkung in der Kulturpflanze Gerste als Reaktion auf Trockenstress. Um weitere Erkenntnisse über die molekularen Mechanismen einer verbesserten Bestockung und Trockentoleranz zu gewinnen, werden zunächst neben der verfügbaren Gerstenmutante hvd14.d, Gerstenakzessionen mit einer natürlichen Variation in der Sequenz des HvD14-Gens physiologisch, morphologisch und zellbiologisch untersucht.
Darüber hinaus werden mittels gezielter Mutagenese (CRISPR RNA / Cas9) und dem Screening einer TILLING-Population weitere Mutanten mit modifizierten Genen der SL-Biosynthese oder des Signalwegs erstellt, um direkte oder indirekte Wechselwirkungen zwischen SLs und ABA aufzuzeigen. Die Tiefensequenzierung differentiell exprimierter Gene, wird durch die Messung der Konzentrationen von SL, ABA, Auxinen, Brassinosteroiden, Cytokininen, Gibberellinen, Jasmonsäure und Salicylsäure mit und ohne Trockenstressbehandlung vervollständigt. Darüber hinaus werden selektierte SL-Mutantenlinien mit ausgewählten Hormonen wie ABA, Brassinosteroiden und Cytokininen behandelt, um vorhergesagte Wechselwirkungen zu bestätigen.

Die Zielsetzung des gemeinsamen Projektes mit der Teilsammlungen Nord der IPK Genbank, Saatzucht Steinach GmbH & Co KG und GenXPro GmbH ist die Entwicklung prakti­kabler Selektionsmethoden für relevante Genotypen die zu einer Verbesserung des Samensitzes und damit der Dreschfestigkeit in züchte­risch bearbeiteten Futter- und Rasengräsern führen. In der Arbeitsgruppe Strukturelle Zellbiologie sollen hierbei mit zellbiologischen Methoden Marker für die indirekte Merkmalsselektion vor der Blüte entwickelt und im Anschluss im Zuchtmaterial validiert werden. Hauptziel war hierbei die eingehende Untersuchung der für den Samenausfall verantwortlichen Abszissionszone auf morphologischer und struktureller Ebene. Für die vergleichenden Analysen in den verschiedenen Entwicklungs­stadien vom Ährenschieben bis zur Fruchtbildung (Samenreife) wurden ausfallfeste und zu Samenausfall neigenden Genotypen herangezogen.  Die Erkenntnisse sollen Aufschluss dar­über bringen, zu welchem Zeitpunkt zwischen den Genotypen eine Differenzierung des Trenn­gewebes sichtbar wird, und den Zeitraum ein­grenzen, in welchem die für die Differenzierung verantwortlichen Gene aktiv sind. Letztendlich sollen die selektierten Genotypen der Entwicklung von Zuchtstämmen dienen, die neben der geforderten Ertragsleistung und Qualität ein deutlich höheres Samenertragspotenzial aufweisen und damit die Saatgutproduktion wesentlich ökonomischer gestalten als bislang.

Die Standfestigkeit ist beim Roggen (Secale cereale L.) wegen seiner gegenüber den anderen Getreidearten großen Wuchshöhe, ein wichtiges Zuchtziel. Der in Roggen identifizierte Genotyp "Stabilstroh" ist durch die Resistenz gegenüber Lager charakterisiert und zeichnet sich zudem durch eine erhöhte Pflanzenlänge und Biomasseproduktion im Vergleich zu anderen deutschen Roggenhybriden aus. Zur Klärung des genetischen Hintergrunds der Lagerresistenz wurde in Kooperation mit den Arbeitsgruppen Ressourcengenetik und Reproduktion, Gen- und Genomkartierung des IPK, sowie dem Züchtungsunternehmen Dieckmann Seeds GmbH, zunächst eine histologische und ultrastrukturelle Charakterisierung der Basal-Internodien der Elternlinien und der segregierenden F2-Population durchgeführt. Mittels der identifizierten Merkmale in Bezug auf die Lagerresistenz, konnten dann 30 genetische Loci (QTLs) auf sieben Chromosomen identifiziert werden. Da zu erwarten ist, dass dieses Merkmal auch zu einer Verbesserung der Standfestigkeit in anderen Getreidearten führen kann, sollen vorselektierte Weizen- und Gerstenakzessionen (Auswertung historischer Felddaten, IPK Genbank), auf die Ausbildung der in Roggen identifizierten Merkmale hin untersucht und charakterisiert werden. In diesem Zusammenhang werden in Kooperation mit den beiden Arbeitsgruppen Metabolische Diversität, Ressourcengenetik und Reproduktion, zurzeit verschiedene Entwicklungsstadien der Elternlinien des Roggengenotyps “Stabilstroh”, eingehend mittels Histologie und Metabolitanalysen untersucht. 

In unserer Funktion als zentrale Einrichtung für Mikroskopie sind wir zudem als Kooperationspartner in die Bearbeitung einer Vielzahl weiterer Projekte eingebunden. 

So wurde unter anderem in den vergangenen Jahren unsere enge Zusammenarbeit mit der Arbeitsgruppe molekulare Pflanzenernährung und CONICET (Rosario, Argentinien) zu einer verbesserten Stresstoleranz von Pflanzen durch die Expression von Flavodoxin (Fld) aus Cyanobakterien fortgesetzt und intensiviert. Es konnte gezeigt werden, dass Fld die verlorene Ferredoxinaktivität unter Stressbedingungen ersetzen kann. Ultrastrukturanalysen bestätigen, dass die Expression von Fld in Tabakpflanzen zu einer Verzögerung des Seneszenzprozesses führte, indem der Stoffwechsel in Blättern, Hormone und Seneszenz assoziierte Gene reguliert wurden. Darüber hinaus zeigt die Ultrastrukturanalyse von Chloroplasten, dass Flavodoxin-exprimierende Tabakpflanzen einem Phänotyp mit hoher Lichtakklimatisierung gleichen.

Für die AG Genomik Genetischer Ressourcen werden unter anderem vergleichende transmissionselektronenmikroskopische Analysen von Chloroplasten von Arabidopsis- und Gerstenmutanten und heterologer Komplementationslinien durchgeführt. Die Ultrastrukturdaten, insbesondere zur Verteilung und Architektur der Grana-Stapel, können hierbei weitere Erkenntnisse zur funktionellen Charakterisierung potentieller Gene der Chloroplastenbiogenese liefern.

In Zusammenarbeit mit der ehemaligen AG Metalloid-Transport konnten eine abnormale Gefäßdifferenzierung als irreversibles Symptom von Bor-Mangels in Raps beschrieben werden. Insbesondere im Xylem und Phloem treten signifikante strukturelle Veränderungen auf, die die Zellstruktur und die Gewebeorganisation beeinflussen. Die morphologischen Veränderungen sind in sensitiven Genotypen deutlich ausgeprägter und unterscheiden sich deutlich von denen, die für andere Nährstoffmängel berichtet werden. Um Gene zu identifizieren, die für die Gefäßdifferenzierung unter B-Mangelbedingungen verantwortlich sind, werden die Forschungsarbeiten mit Prof. Patrick Bienert von der TU München fortgesetzt. Vorläufige Daten aus der RNA-Sequenzierungsanalyse, einer Analyse zur Genontologie und der Identifizierung differentiell exprimierter Gene legen hierbei eine Neuorganisation des Metabolismus und Differenzierungsprozesse unter B-Mangelbedingungen nahe.

Neben einer Reihe von internen und externen Kooperationen werden in Zusammenhang mit Forschungsarbeiten der unabhängigen AG Pflanzliche Baupläne umfassende zellbiologische Untersuchungen zur Ährchen- und Blütchenbildung unterschiedlicher Entwicklungsstadien von Weizen und Gerste durchgeführt. Darüber hinaus wurde die Erstellung eines umfassenden histologischen Atlas zur Gefäßarchitektur der zweireihigen Gerstenähre während ihrer Entwicklung begonnen. Anhand von Schnittserien und 3D-Modellen soll die Organisation der vaskulären Gefäße in den Nodien, sowie zwischen Rachis und den Ährchenorganen, visualisiert werden.

Mehrere Kooperationen mit der AG-Chromosomenstruktur und -funktion befassen sich mit der ultrastrukturellen Charakterisierung von Zellkernen. Basierend auf Hochdruckgefrieren für die Probenpräparation konnten wir zeigen, dass sich die Chromatinstruktur und -replikation zwischen Mikrokernen und Primärkernen unterscheidet. Die Ultrastrukturanalysen stützen hierbei weitere Daten zur genauen Eliminierung überzähliger B-Chromosomen von Aegilops speltoides in Wurzeln zu Beginn der Embryoentwicklung (Ruban et al.,. 2020, Nat Commun.).
