Entwicklungs- und Molekulargenetik von Getreide:
Das Forschungsprogramm konzentriert sich hauptsächlich auf die Ertragsbildung bei Weizen und Gerste. Wir verwenden natürlich vorkommende Variationen von Weizen und induzierte Mutanten von Gerste, um die genetischen Zusammenhänge von Genen zu klären, die sog. Entwicklungsphänotypen zugrunde liegen, wie z.B. für reduzierte und erhöhte Kornzahl pro Ähre.

Projekte

Ein vielversprechender Weg zur Verbesserung des Ertrags von Getreidekulturen, einschließlich Weizen und Gerste, ist die Verringerung der Sterblichkeit von Ährchen/Blütchen. Ährchen, die körnerproduzierenden Organe von Getreideähren, bilden sich normalerweise im Überschuss und abortieren anschließend während der Entwicklung. Ein erhöhtes Überleben von Ährchen/Blütchen ist mit einer erhöhten Anzahl von Körnern pro Ähre verbunden. Daher ist die Verringerung der Sterblichkeit von Ährchen / Blütchen ein faszinierender Ansatz zur Verbesserung des Kornertrags pro Ähre.

Bei Gerste repräsentiert die Anzahl der Ährchen pro Ähre im Stadium um das Grannenprimordium das maximale Ertragspotential pro Ähre. Danach führt eine signifikante Ährchensterblichkeit zu weniger Körnern. Unsere früheren Ergebnisse zeigten, dass das Überleben von Ährchen/Blütchen in Gerste stark genetisch kontrolliert wird (Alqudah und Schnurbusch 2014, Functional Plant Biology). Die zugrunde liegenden genetischen und molekularen Determinanten des Überlebens der Ährchen müssen jedoch noch entdeckt werden. Die im Rahmen des Projekts LUSH SPIKE (ERC CoG, 2016-2021) erzielten Ergebnisse werden daher unser Verständnis für die Verbesserung der Erträge von Getreide verbessern.

Blütenstandsarchitektur von Getreideähren

Unser Verständnis der Molekulargenetik der Ähren- oder Ährchenentwicklung ist bei Getreidepflanzen sehr begrenzt. Funktionelles Wissen über Gene, die wichtige Entwicklungsmerkmale wie Blütenstandsverzweigung, Ährchenformation oder -abort, Länge der Spindelinternodien oder die Gesamtzahl der Internodien der Spindeln regulieren, fehlt in den meisten unserer Getreidekulturen fast vollständig. Wir verwenden natürliche Ährenvarianten aus Weizen und induzierte Ährenmutanten aus Gerste, um die genetische Ausstattung von Genen zu klären, die Entwicklungsphänotypen für eine reduzierte und erhöhte Körnerzahl pro Ähre zugrunde liegen.

Ährchen- und Blütchenentwicklung

Bei den Triticeae zeigt die Gerste (Hordeum vulgare L.) eine charakteristische Ährchenbildung mit drei einblütigen Ährchen pro Spindelstufe. Aufgrund der Größe und Fruchtbarkeit der beiden seitlichen Ährchen kann die Gerste in fünf Formen eingeteilt werden (Mansfeld 1950): (1) convar. deficiens, (2) konvar. distichon, (3) konvar. intermedium, (4) konvar. labile und (5) konvar. hexastichon. Drei Gersten-Loci – vrs2, vrs3 und vrs4 – bestimmen jeweils die Umwandlung von sog. zweizeiliger (Distichon) zu sechszeiliger (Hexastichon) Gerste, wurden jedoch ausschließlich nach Mutagenese gefunden. Die molekulare Isolierung von Genen, die an der Fruchtbarkeit der seitlichen Ährchen beteiligt sind, kann einen wichtigen Beitrag zu weiteren Erkenntnissen über die genetischen Entwicklungsprozesse liefern, die der Blütchenbildung bei Gerste und anderen Gräsern zugrunde liegen.

Die zugrunde liegenden genetischen und molekularen Determinanten des Überlebens von Ährchen müssen jedoch noch entdeckt werden. Daher werden die Ergebnisse des Projekts LUSH SPIKE (ERC CoG, 2016-2021) unser Verständnis davon verbessern, wie die Erträge von Getreidepflanzen verbessert werden können.
